## What we do

The Baron-Gori Lab focuses on understanding the molecular, cellular, and genetic mechanisms behind bone remodeling and skeletal homeostasis in both animal models and humans. We utilize primary cells, cell lines, and mouse genetics, particularly studying conditions such as osteoporosis, lactation, rare diseases, and fracture repair.

### Research areas

#### Wnt signaling and bone

Wnt signaling is a critical pathway in bone development and regulation of bone mass. Our research has contributed to developing therapies, such as sclerostin inhibitors for severe osteoporosis. We explore Wnt pathway components, including recent studies on Sfrp4, which influence bone regeneration and skeletal homeostasis. Dr. Gori’s work, supported by an NIH grant, delves into the role of Sfrp4 in cortical bone formation.

#### Osteocyte function in bone remodeling

Osteocytes, the most abundant bone cells, regulate bone remodeling through sclerostin and RANKL, which are key to treating osteoporosis. Our research examines their ability to resorb and form bone matrix, known as osteocytic osteolysis and perilacunar apposition, affecting calcium metabolism, particularly during lactation.

#### PTH/PTHrP signaling and skeletal homeostasis

PTH is an anabolic drug used in osteoporosis treatment. Our studies explore its role in reducing bone marrow fat and increasing bone formation, focusing on transcription factors like Zfp521, which promote bone formation and inhibit marrow fat. These findings are part of NIH-funded projects investigating PTH’s mechanisms in bone and fat regulation, particularly in aging individuals.

This structured focus enables us to investigate key pathways and develop therapeutic strategies for both common and rare skeletal diseases.
